LC Columns and AccessoriesLC Column SelectionInformation in the following section will help you make an informed decision on theappropriate HPLC column for your application, based on stationary phase use, analyteproperties, LC-MS requirements or USP specifications. You will also find a useful tableof Thermo Scientific phases with specifications, as well as recommended ThermoScientific alternatives for other popular columns.Refer to the Advanced User Graphic (AUG) on thecorresponding product page (illustrated to the right)for more help and information on general purposecolumn selection. The AUG will show you Hydrophobicity which givesthe relative retention on the column. Generally, thehigher the hydrophobicity, the greater the retentionof neutral compounds and the higher the organiccontent in the mobile phase. A lower value indicatesa need for higher aqueous mobile phases to achievecomparable retention and resolution. Therecommended pH Range for the column is illustrated, outside of which column lifetimes will diminish. The Pore Size is shown, with larger pore size columnsbeing more applicable to larger analytes such asproteins or peptides. The percentage Carbon Loadis related to the hydrophobicity. Below the icon, youwill see the particle sizes available, as well as theUSP code. These graphics are designed to allow youto quickly compare the main characteristics of multiplestationary phases, allowing you to choose quickly themost appropriate stationary phase for your analysis. For additional help in column selection, pleasesee the back cover to contact our expert TechnicalSupport and tap into our expertise to help makethe best choice for your application.HydrophobicityLowHighpH Range02 to 814Pore Size080Å300Carbon Load (%)05Particle Size 2.6µm25USP L11Advanced User Graphic (AUG)Thermo Scientific Chromatography Columns and Consumables 2016-2017Common HPLC Phases and Their ilicaSilica-OHC1SAS-(CH3) 3 Least retentive of all alkyl group bonded phases for non-polar solutes.Typically used for moderately polar and multi-functional compounds.C4Butyl-C 4H9 Shorter retention than C8, C18. Separation of peptides and proteins.C8MOS-C 8H17 Less retentive than C18; normally used for small peptides andproteins, pharmaceuticals, steroids, environmental samples.C18ODS-C18H37 Most retentive of the alkyl-bonded phases. Used widely forpharmaceuticals, steroids, fatty acids, phthalates, environmental etc.CyanoCPS, CN-(CH2) 3CN Unique selectivity for polar compounds, more suitable than basesilica for normal phase gradient separations. When used in reversedphase, the selectivity is different to that of the C8 and C18 phases.Useful for a wide range of pharmaceutical applications and formixtures of very different solutes.
